Abstract
Rats serve as important reservoirs for a wide range of diseases. Rapid urbanization leads to their infestation near human enclosures, which is a major source of zoonotic RNA virus transmission from rats to people. Members of the Paramyxoviridae family have tropism for the upper and lower respiratory systems, causing respiratory problems in humans and rodents. Paramyxoviridae virus, such as the Mojiang virus in rats, can also infect humans. Although paramyxoviruses have been detected in rats in other Asian countries like China and Vietnam, however, none has been screened in Malaysia. Thus, this study aims to conduct a preliminary screening to detect the presence of Paramyxoviridae RNA in rats in Klang Valley, Selangor, Malaysia. A total of 35 rats were collected from several markets in Klang Valley for blood serum and oropharyngeal swabs collection. The RNA of the samples were extracted using TRIsureTM reagent, followed by a semi-nested reverse transcription-polymerase chain reaction (RT-PCR) targeting the polymerase (pol) gene and yielding a 561 bp product and lastly gel electrophoresis on 2.0% agarose gel. The reactions were then run on 2.0% agarose gel. There was no discernible band to positive controls indicating Paramyxoviridae RNA was not detected in all samples of this study. The negative results suggested that the rats were not currently infected with the viruses but did not indicate they had no prior exposure. It might, however, be attributable to the limited sample size and sample types, which should be enlarged and expanded for further detection of Paramyxoviridae in rats.
