Abstract

This study was conducted to evaluate the antibacterial properties and cytotoxic activities of different parts of two Acacia species. In this research, crude extracts of phyilodes, stem, and seed of Acacia mangium and crude extracts of phyllodes, stem, seed, and flower of Acacia auriculiformis were tested on four strains of microbes which were Escherichia coli, Vibrio parahaemolyticus, Listeria monocytogenes, and Pseudomonas aeruginosa for antimicrobial activity by using Disc Diffusion Method. For brine shrimp lethality assay, all of these extracts were tested on brine shrimp nauplii (Artemia salina) to screen their cytotoxicity. All of the tests were done in triplicate. From the study of antimicrobial properties, all of the extracts showed positive results. The leaf of Acacia auriculiformis showed good antibacterial activity against all four strains of microbes where the extract can inhibit all of the strains at concentration levels of 125 mg/ml. Among all four strains, E. coli exhibited the highest rates of inhibited growth when tested with the crude extracts of different parts of both Acacia species compared to the other three strains as this strain was inhibited by all of the extracts with almost all showing the highest inhibition zone. For all extracts used in this assay, the lowest level of concentration that can inhibit the growth of E. coli, V. parahaemolyticus, L. monocytogenes, and P. aeruginosa was at 125 mg/ml. For cytotoxic activity, the crude seed extract of Acacia auriculiformis showed the strongest activity with LCso 81.1 µg/ml. The crude phyllodes extract of A. auriculiformis however showed most negative result where this extract could not kill more than 50% of A. salina nauplii. In general, based on the results, the crude extracts of Acacia species used in this study had more positive to antibacterial properties in this assay compared to brine shrimp lethality assay.
