Abstract
The objective of the present study was to isolate and identify bacteria from acid soil, Bekenu series using morphological, biochemical and molecular techniques. Soil samples were collected from regions of Universiti Putra Malaysia Bintulu Sarawak Campus (UPMKB) in a simple random technique. A total of 11 soil bacteria samples were isolated and 7 of them could be molecularly identified. The rest 4 couldn't be identified due to time constrain. Morphological shapes of colonies were initially used to separate the grown bacteria of the samples. These bacterial isolates were Gram negative coccoid rod, Gram negative rods and Gram positive rods. Twelve biochemical tests were performed to study their biochemical characteristic. The test conducted are skim milk agar test, cellulose test, sprit blue agar test, starch agar test, Eosin Methylene Blue (EMB) agar test, Violet red bile agar test, MacCONKEY agar test, gelatin liquefaction test, Klinger Iron agar test, Catalase test and Lactose peptone broth test. Molecular technique comprises of genomic DNA extraction, polymerase chain reaction (PCR) amplification of 16S rRNA, DNA purification, gel electrophoresis and DNA sequencing were performed. DNA sequencing results were analyzed using online database Basic Local Alignment Search Tool (BLAST). Based on the 16S rRNA gene sequencing bacteria identified belongs to two major groups, Bacilli and B-proteobacteria. They were identified as Chromobacterium violaceum, Cupriavidus basi/ensis, Bacillus subtilis, Bacillus cereus, Bacillus pycnus and Bacillus thuringiensis. These studies demonstrate the great diversity of bacteria in acid soil.
