Abstract
Leishmaniosis is a vector-borne disease caused by obligate intracellular protozoa of genus Leishmania. Leishmania infantum is the most common and important cause of canine leishmaniosis worldwide. There has been no reported case of canine leishmaniosis in Malaysia however two human leishmaniosis cases was reported in Malaysia on 1995 and 2007 respectively. Veterinarians from private clinics have claimed to treat dogs showing similar clinical signs to canine leishmaniosis. To detect the presence of Leishmania sp. in dogs‟ population in Malaysia, blood samples were taken from dogs in animal shelters in Sabah and Sarawak. Fifty four blood samples were taken from Sabah and 50 blood samples were taken from Sarawak. The blood was taken from the cephalic vein aseptically and subjected to DNA extraction kits and conventional PCR using the primers targeting the L. infantum kinetoplastminicircle DNA. Amplification products were analyzed by electrophoresis on agarose gels in 1.5x Tris-acetate EDTA buffer. Out of 54 samples from Sabah, 31 appeared positive on PCR with the prevalence of 57.4%, meanwhile in Sarawak, 33 out of 50 samples appeared positive on PCR with the prevalence of 66%. Eight positive samples from each state were subjected to sequencing. The presence of L. infantum in Malaysia could be due to presence of the vector, sandfly which inhabits the cave area which is a lot all over Sabah and Sarawak. This is the first detection of canine leishmaniosis in Malaysia.
