Abstract
Imidazole and benzimidazole are important pharmacophore in the drug discovery process as they display a variety of pharmacological activities. Benzimidazole and its derivatives possess various pharmacological activities such as antimicrobial, antiviral, anticancer, antiprotozoal, anti-inflammatory as well as antioxidant. Objective: This study was done to evaluate the antioxidant capacity of the two benzimidazole derivatives which are (E)-4-(5,6-dimethyl-1H-benzo[d]imidazol-2-yl-N’-3-hydroxybenzylidine)benzohydrazide (Compound NN-1-5) and (E)-N’-(2,5-dihydroxybenzylidine)-4-5,6-dimethyl-1H-benzo[d] imidazol-2-yl) benzohydrazide (Compound NN-1-7) by using different antioxidant assays. This study aims to compare the difference in antioxidant activities of the compounds by using DPPH radical scavenging assay, FRAP assay and PAP assay. Method: In this study, the antioxidant activity was determined by the analysis of DPPH radical scavenging antioxidant assay , ferric reducing antioxidant power (FRAP) assay and phosphomolybdenum antioxidant power (PAP) assay. The concentration is fixed between 3.125 μM - 200 μM for FRAP assay as well as 6.25 μM - 400 μM for DPPH. Result: The results from the DPPH and FRAP antioxidant assay shows that Compound 7 exhibits good antioxidant properties as compared to Compound 5 and the standard used. For DPPH assay, the EC50 of Compound 7 is 35.51 μg/ml, while the highest concentration of Compound 5 does not yield 50% of the DPPH radical scavenging activity. As for FRAP assay, the antioxidant activity of compound 7 is 98.083 mmol Fe2+/g which is slightly higher than compound 5. Conclusion: The benzimidazole derivative which is Compound 7 has been proved to poses better antioxidant properties compared to Compound 5.
