Abstract
A study was carried out to determine the genetic status of red tilapia populations cultured at UPMKB using microsatellite markers and to assess the potential benefits of using existing populations for future breeding programmes. 40 red tilapia specimens were collected from two locations (pond culture system and tank culture system). 4 microsatellite markers were used for further fragment analysis. All loci showed 100% polymorphism in both populations. The number of alleles per locus ranged from 3 to 8. The effective number of alleles (Ne) ranged from 2.480 to 2.737. The observed heterozygosity (Ho) and expected heterozygosity (He) ranged from 0.481 to 0.563 and 0.548 to 0.612, respectively. Shannon information index for the tank and pond populations were 0.983 and 1.127 respectively. Fixation index for the tank and pond populations are 0.127 and 0.08 respectively. FST values ranged between 0.002 (GM128) and 0.045 (GM047), with a mean of 0.016. The mean number of migrants per generation (Nm) was 55.647. The distribution of genetic variation within individuals was high (83%). There were no significant differences between tank and pond populations. The recommendation for further breeding programmes is to look for red tilapia with high genetic diversity.
