Abstract

Countless methods have been investigated throughout the years to determine the quickest and most reliable way to assess sperm quality. One of the prominent methods would be to perform MTT (3-(4, 5-dimethylthiazol-2-y1)-2, 5-diphenyltetrazolium bromide) assay reduction on metabolically active sperm cells. The purpose of this study is to investigate whether MTT assay is a good way to assess quality of bovine sperm which will be beneficial in the cattle industry especially for artificial insemination. In this study, 3 fresh and 4 frozen-thawed semen samples obtained through electroejaculation were used for MTT assay. Semen evaluation was performed to check for motility, live sperm percentage, wave pattern and concentration before proceeding with MTT assay. Using Pearson correlation coefficient analysis, MTT absorbance rates increased significantly (P<0.05) as the live sperm percentage increases. There was a high positive correlation (r= 0.8371, P<0.05) between MTT absorbance rates and sperm viability. Same tests were repeated to check for correlation between MTT absorbance rate and semen evaluation parameters like motility, wave pattern and as a result, there is positive correlation with wave pattern r=0.8694 (P<0.05) but test is not significant for motility (P>0.05). In summary, MTT assay is a reliable method for assessment of bovine sperm quality to enhance efficiency and productivity of the cattle industry
