Abstract
Flaviviruses are mostly arthropod-borne viruses (arboviruses) of medical and veterinary field significance. Many different flavivirus species have been isolated from various hosts including humans, animals, ticks and mosquitoes. Studies have shown that wild rats could act as reservoirs of many zoonotic pathogens with or without vector involvement. Since there is a lack of study on the prevalence of Flavivirus in Rattus spp. in Malaysia, this study therefore was aimed to detect Flaviviridae among rats in Klang Valley, Malaysia. Thirty five wild rats were captured at several wet markets in Klang Valley. Prior to getting euthanized through cervical dislocation, the rats were anesthetized by chloroform. Blood serum and oropharyngeal swabs were taken for molecular analysis. RNA extraction using TRIzol followed by a one step reverse transcriptase-polymerase chain reaction (RT-PCR) assay targeting the NS5 gene was performed. All 70 samples tested were negative for Flaviviridae since there were no specific bands formed following electrophoresis. Although none of the tested samples were positive, the risk of zoonotic transmission of flavivirus should still be taken into consideration as it can have the potential to pose a public health threat.
