Abstract
Bario rice, which is famous for its quality and unique taste, is originally comes from the highlands of Sarawak. Nowadays, there is an increasing demand for Bario rice causing a leap in the price of this kind of rice. In relation to this, there is an issue to authenticate the originality of Bario rice that is sold under this name. The objectives of this study were (i) to authenticate different "claimed" Bario rice variety: Bario rice accession from one paddy growing area in Bario, and Bario rice accession from the local seller in Bintulu using RAPD-PCR method, (ii) To compare the banding pattern of Bario rice genetic sequence with other variety of rice : Aromatic Thai rice. Bario rice of Adan variety from Pa' Lungan was used as a standard match. Simple DNA extraction method presented in this study is based on a freeze-and-thaw cycle that enables extraction of purified high molecular weight grain DNA without the use of expensive equipment and harmful reagents. Next, DNA samples were subjected to RAPD-PCR analysis. This technique was chosen because RAPD-PCR uses single 10-mer arbitrary primers that requires much less DNA, and is technically simpler and cheaper compared to other PCR-based technique. We were unable to meet the objectives of this study because of two main obstacles that we faced during the commencing of this research (i) the quality and yield of our DNA extraction product which is questionable due to the inconsistency of the spectrophotometer reading (ii) limited time available to optimize the specific and suitable PCR cocktails mixture with the right concentration and PCR thermocycling profiles especially the annealing temperature because only a specific temperature profile works for difference DNA sequence.
