Abstract
Introduction: Plant based products (PBP) consist of plant food supplement (PFS) and herbs are commonly used by the consumers without knowing the actual ingredients in the PBP and they thought that all "natural" ingredients are "safe". Aristolochic acids (AA) is a genotoxic and carcinogenic compound that is naturally exist in plant such as wild ginger and birthworth. It is well known to be the genotoxic and has been classified in Group 1 carcinogenic. Objective: This study was conducted to determine the risk from exposure to AA via consumption of PBP using margin of exposure (MOE) approach. Methodology: Ten local PBP were purposively purchased and extracted using methanol extraction for PFS and aqueous extraction for herbs. The level of AA in the samples were then quantified using Ultra High Performance Liquid Chromatography (UHPLC). MOE approach was applied to calculate the risk for both genotoxic and carcinogenic substance by diving the Benchmark dose level (BMDL10) with estimated daily intake (EDI). Results and discussion: Four out of ten samples were detected to contain AA. The highest in sample one with 3.22 (±2.52) mg/g and the lowest from sample nine, 1. 78 (±0.10) mg/g. The EDI of AA from the consumption of PBP ranging from 1.14 x 10-1 to 4.63 x 10-1 mg/kg bw/day. MOE was then calculated using lowest BMDL10 which is 10 mg/kg bw/day and all four positive samples have MOE less than 10,000. Sample one has the lowest MOE with 2.16 x 10- 2 and the highest MOE value, 8.79 x 10-1 was from sample nine. Conclusion: All positive samples required high priority for risk assessment since the value for MOE less than I 0,000.
