Abstract

In this research, glucoamylase was immobilized onto sodium alginate and cellulose nanoparticle which is known as cellulose that have properties as plant cellulose. The commercial glucoamylase enzyme from Aspergillus sp. Was immobilized onto sodium alginate and cellulose nanoparticle through modified entrapment technique. Protein loading onto the support was analysed by using UV NIS Spectrophotometer and the enzymatic activity of producing glucose was analysed by HPLC-Ri. Based on the entrapment method of glucoamylase onto the sodium alginate support only resulted about 33.8% of enzyme loading with 10.8% of enzyme acitivty. Then, for the immobilization of glucoamylase onto the sodium alginate and cellulose nanoparticle support resulted about 62. l 0% of enzyme loading with 45.62% of enzyme activity. Next, the reusability of immobilized glucoamylase was also analyzed by using same immobilized glucoamylase for 6 cycles of reaction and the product was tested by using HPLC-Ri. For immobilized enzyme that use sodium alginate only, it resulted in 50% activity retained while for immobilized enzyme that use both support which is sodium alginate and cellulose nanoparticle, it resulted in 70% activity retained. Besides that, the surface morphology study was carried out by using Scanning Electron Microscopy (SEM), the difference in surface morphology of immobilized glucoamylase for with and without cellulose nanoparticle was obtained.
